Cytotoxicity Evaluation of A New Resin Matrix System for Dental Fiber-Reinforced Composites

Abstract
Objectives. The purpose of this study was to determine the cytotoxicity of HDMA-based E-glass FRCs. Methods and Materials. The materials used were bis-GMA (Sigma-Aldrich, USA), MMA (ProSciTech, Australia), HDMA (Esstech, USA), CQ (Esstech, USA), CEMA (Esstech, USA), E-glass fibers (Ahlstrom, Finland) and fibroblast cell line (UGM, Indonesia). Fifteen FRCs (2.0mmx2.0mmx25mm) were prepared and divided into 3 groups. The experiment groups were 78.4wt%HDMA+19.6wt%MMA+1.0wt%CQ+ 1.0wt%CEMA (Exp1-group) and 49.0wt%HDMA+49.0wt%MMA+1.0wt%CQ+ 1.0wt%CEMA (Exp2- group). The Control-group was 78.4wt%bis-GMA+19.6wt%MMA+ 1.0wt%CQ+1.0wt%CEMA. The specimens were milled, diluted in a culture medium, contacted to the cells for 24 h then evaluated by MTT method. Cells viability was analyzed by ANOVA and LSD. Result. The average of cells viability in percentage showed for the Exp1-group was 67.73±0.71, Exp2-group was 66.78±0.20 and Control-group was 64.36±1.63. Analysis by ANOVA revealed significant difference of cells viability among the groups (p<0.01). The LSD test showed significant difference between the Exp1-group and Control-group and between the Exp2-group and Control-group (p<0.01); whilst no significant difference between the Exp1-group and Exp2- group (p>0.05). Conclusions. An experimental HDMA-based E-glass FRCs (the Exp1-group and Exp2-group) revealed significant difference on cells viability with bis-GMA-based E-glass FRCs. The HDMA-based E-glass FRCs was less cytotoxic than bis-GMA-based E-glass FRCs.
