Karena sumbernya relatif mudah diakses (melalui ekstraksi gigi atau sisa pulpa), kemampuan diferensiasi yang luas, dan potensi regeneratif, DPSCs menjadi salah satu kandidat utama dalam penelitian regenerasi oral dan implantologi gigi.<\/p>\n\n\n\n<p>Melalui penggunaan metabolit dari DPSCs dalam perancah biomaterial, penelitian terkini menunjukkan bahwa DPSCs tidak hanya berperan sebagai sel asli yang berdiferensiasi, tapi juga berfungsi melalui sekretoma \/ metabolitnya dalam memodulasi lingkungan jaringan sekitarnya.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Metabolit DPSCs &amp; Ekspresi COL1A1<\/strong><\/h2>\n\n\n\n<p>Penelitian oleh mahasiswa FKG UGM, <strong>EUGENIUS BRAMIANTA WARDHANA, dibimbing oleh drg. Murti Indrastuti, M.Kes., Sp.Pros(K)&nbsp; dan drg. Intan Ruspita, M.Kes., Ph.D., Sp.Pros(K)<\/strong><em>&nbsp; <\/em><strong>dengan judul<\/strong><em> \u201cPengaruh Pemberian Metabolit Sel Punca Pulpa Gigi pada Perancah Gelatin terhadap Ekspresi Protein COL1A1 pada Proses Oseointegrasi Implan Gigi\u201d<\/em> menguji bagaimana metabolit dari DPSCs apabila dipadukan ke dalam scaffold gelatin dapat memengaruhi ekspresi gen\/protein <strong>COL1A1<\/strong> (kolagen tipe I) dalam konteks integrasi implan gigi ke dalam jaringan tulang. Beberapa poin utama dari penelitian tersebut:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>DPSCs dikultur dan metabolit yang dihasilkan diambil, lalu diaplikasikan ke scaffold gelatin yang ditanam di dekat implan gigi dalam model penelitian.<\/li>\n\n\n\n<li>Hasil menunjukkan bahwa pemberian metabolit DPSCs pada scaffold gelatin bisa meningkatkan ekspresi protein COL1A1 selama proses oseointegrasi implan gigi, yang penting karena kolagen tipe I adalah komponen utama matriks tulang.<\/li>\n\n\n\n<li>Temuan ini mendukung ide bahwa DPSCs bekerja tidak hanya melalui diferensiasi sel-sel baru, tetapi juga melalui efek parakrin (sekresi metabolit\/faktor pertumbuhan) yang memfasilitasi pembentukan jaringan tulang di sekitar implan. (Penelitian di UGM)<\/li>\n<\/ul>\n\n\n\n<p>Dengan demikian, penelitian ini memperluas perspektif fungsi DPSCs dalam regenerasi jaringan oral \u2014 bukan hanya \u201cmenjadi sel tulang baru,\u201d tetapi juga \u201cmengatur lingkungan jaringan\u201d melalui metabolit.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Karakteristik DPSCs dan Mekanisme Kerja<\/strong><\/h2>\n\n\n\n<ol class=\"wp-block-list\">\n<li><strong>Sumber dan Isolasi<\/strong><br>DPSCs diperoleh dari pulpa gigi yang sehat, paling sering pada gigi yang harus diekstraksi (misalnya gigi geraham bungsu). Pulpa diambil, dikultur dalam medium khusus, dan DPSCs diperoleh melalui isolasi subpopulasi yang menunjukkan sifat stemness (marker mesenkimal, kemampuan proliferasi).<\/li>\n\n\n\n<li><strong>Potensi Diferensiasi<\/strong><br>DPSCs mampu berdiferensiasi menjadi osteoblas (tulang), kondrosit (kartilago), adiposit (lemak), sel saraf, dan pulpa regeneratif.<\/li>\n\n\n\n<li><strong>Sekretoma \/ Metabolit Parakrin<\/strong><br>DPSCs menghasilkan faktor-faktor pertumbuhan, sitokin, dan molekul sinyal lainnya dalam medium kultur (metabolit). Metabolit ini dapat mempengaruhi sel-sel di sekitarnya: merangsang proliferasi, migrasi, angiogenesis, dan matriks ekstraseluler, termasuk kolagen tipe I (COL1A1).<\/li>\n\n\n\n<li><strong>Peran dalam Oseointegrasi Implan<\/strong><br>Dalam konteks penelitian kutipan, metabolit DPSCs membantu meningkatkan ekspresi COL1A1 di sekitar implan, yang sangat penting untuk pembentukan dan integrasi matriks tulang baru di permukaan implan.<\/li>\n<\/ol>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Aplikasi Klinis dan Potensi<\/strong><\/h2>\n\n\n\n<ul class=\"wp-block-list\">\n<li><strong>Implantologi Gigi<\/strong>: Penggunaan DPSCs (atau metabolitnya) dapat mempercepat integrasi tulang di permukaan implan, meningkatkan stabilitas awal dan jangka panjang.<\/li>\n\n\n\n<li><strong>Regenerasi Tulang Alveolar<\/strong>: DPSCs bisa digunakan dalam sayatan bedah dan pembentukan graft tulang di daerah defek alveolar.<\/li>\n\n\n\n<li><strong>Terapi Luka Oral &amp; Pulpa Regeneratif<\/strong>: Karena kemampuannya mempercepat regenerasi dan meningkatkan matriks kolagen, DPSCs bisa dipakai dalam terapi luka atau revitalisasi pulpa.<\/li>\n\n\n\n<li><strong>Scaffold Biokompatibel dengan Metabolit<\/strong>: Kombinasi scaffold (misalnya gelatin) dengan metabolit DPSCs (seperti dalam penelitian) menjadi pendekatan biomaterial aktif untuk regenerasi jaringan.<\/li>\n<\/ul>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Challenges and Considerations<\/strong><\/h2>\n\n\n\n<ul class=\"wp-block-list\">\n<li>Stabilitas, dosis, dan metode pelepasan metabolit DPSCs dalam scaffold perlu distandarisasi agar efeknya optimal dan konsisten.<\/li>\n\n\n\n<li>Keamanan jangka panjang, termasuk kemungkinan stimulasi jaringan abnormal (fibrosis, kalsifikasi berlebihan) harus dipastikan melalui penelitian pra-klinis dan klinis.<\/li>\n\n\n\n<li>Translasi dari model laboratorium ke aplikasi manusia memerlukan uji klinik yang hati-hati dan regulasi yang ketat.<\/li>\n<\/ul>\n\n\n\n<p class=\"has-text-align-center\"><strong>*<\/strong>**<\/p>\n\n\n\n<p><strong>Dental Pulp Stem Cells (DPSCs)<\/strong> adalah sumber sel punca oral yang menjanjikan untuk aplikasi regeneratif, baik melalui diferensiasi langsung maupun efek parakrin melalui metabolitnya. Penelitian di UGM menunjukkan bahwa metabolit DPSCs yang diaplikasikan dalam scaffold gelatin mampu meningkatkan ekspresi <strong>COL1A1<\/strong>, mendukung pembentukan matriks kolagen pada proses oseointegrasi implant gigi.
